چکیده

Kearns-Sayre syndrome is a mitochondrial disease, presenting findings before the age of 20 and characterized by chronic progressive external ophthalmoplegia and pigmentary retinal degeneration. It affects many organs, resulting in a very wide spectrum of complications. In this work, a 24-year-old female, whose complaints first started at the age of 12, showing progressing external ophthalmoplegia and diagnosed with Kearns-Sayre disease following mitochondrial changes in muscle electron microscope investigation was presented. Ways of diagnosing in mitochondrial diseases, especially those in children were tried to be evaluated. (J Int Dent Med Res 2010; 3: (3), pp. 154-157)

Progressive external ophthalmoplegia (PEO) due to a mutation in the C10orf2 (PEO1) gene mimicking a myasthenic crisis.

We described a case of a patient with autosomal dominant progressive external ophthalmoplegia (PEO) who presented with the acute onset dysphagia, quadriparesis, ptosis and respiratory insufficiency following a cardiac procedure and mimicking a myasthenic crisis. A pathogenic mutation in the C10orf2 (PEO1) gene was confirmed. The unusual presentation of our patient contributes to expand the clin...

Clinical and molecular features of POLG-related mitochondrial disease.

The inability to replicate mitochondrial genomes (mtDNA) by the mitochondrial DNA polymerase (pol γ) leads to a subset of mitochondrial diseases. Many mutations in POLG, the gene that encodes pol γ, have been associated with mitochondrial diseases such as myocerebrohepatopathy spectrum (MCHS) disorders, Alpers-Huttenlocher syndrome, myoclonic epilepsy myopathy sensory ataxia (MEMSA), ataxia neu...

Levator palpebrae biopsy and diagnosis of progressive external ophthalmoplegia.

BACKGROUND Progressive external ophthalmoplegia (PEO) is a mitochondrial myopathy of ocular muscles. Diagnostic investigation usually involves limb skeletal muscle biopsy and molecular genetic studies, although diagnostic yield tends to be low. The purpose of this study was to evaluate the diagnostic yield obtained by analysis of levator palpebrae (LP) muscle tissue. METHODS This is a clinico...

Association of novel POLG mutations and multiple mitochondrial DNA deletions with variable clinical phenotypes in a Spanish population.

BACKGROUND Both dominant and recessive mutations were reported in the gene encoding the mitochondrial (mt) DNA polymerase gamma (POLG) in patients with progressive external ophthalmoplegia (PEO). Phenotypes other than PEO were recently documented in patients with mutations in the POLG gene. OBJECTIVE To screen patients with mitochondrial disease and multiple mtDNA deletions in muscle for muta...

Two novel mutations in PEO1 (Twinkle) gene associated with chronic external ophthalmoplegia☆

Maintenance and replication of mitochondrial DNA require the concerted action of several factors encoded by nuclear genome. The mitochondrial helicase Twinkle is a key player of replisome machinery. Heterozygous mutations in its coding gene, PEO1, are associated with progressive external ophthalmoplegia (PEO) characterised by ptosis and ophthalmoparesis, with cytochrome c oxidase (COX)-deficien...
